Product Specific Information

Description:
The 24-31 monoclonal antibody reacts with human CD154, also known as gp39 or CD40 ligand (CD40L), a 39 kDa transmembrane glycoprotein belonging to the TNF superfamily. CD154 is transiently expressed by activated T cells and binds to CD40 on antigen-presenting cells (APCs) such as B cells, monocytes/macrophages, and dendritic cells. This interaction is essential for T-dependent B-cell activation and proliferation. Clone 24-31 cross-reacts with cynomolgus monkey CD154.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of stimulated normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test, defined as staining a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD40 Ligand (CD40-L or CD154) is a membrane glycoprotein expressed on activated T cells. It stimulates B cell proliferation and immunoglobulin secretion in the presence of cytokines and induces cytokine production and tumoricidal activity in monocytes. CD40 Ligand also co-stimulates activated T cell proliferation, leading to IFN-γ, TNF-α, and IL-2 production.
