Target Information
Methylation of DNA at cytosine residues in mammalian cells is a heritable, epigenetic modification essential for gene expression regulation, genomic imprinting, and development. Mammalian DNA methyltransferases include DNMT1, DNMT2, and DNMT3 families.
- DNMT1: Maintenance methyltransferase, active in proliferating cells.
- DNMT3A / DNMT3B: De novo methyltransferases, highly expressed in embryonic stem cells.
- DNMT2: Low expression, minimal role in methylation maintenance.
- DNMT3L: Catalytically inactive regulator of DNMT3A/B, expressed in embryonic stem cells, testis, ovaries, and thymus. Forms heterotetrameric complexes with DNMT3A/B. Contains an amino-terminal ADD domain and a carboxy-terminal methyltransferase-like domain.
