Target Information

ATP6V0A1 encodes a component of vacuolar ATPase (V-ATPase), a multisubunit enzyme that mediates acidification of eukaryotic intracellular organelles.
V-ATPase–dependent organelle acidification is essential for intracellular processes such as:

  • Protein sorting
  • Zymogen activation
  • Receptor-mediated endocytosis
  • Synaptic vesicle proton gradient generation

V-ATPase consists of:

  • V1 domain (cytosolic): Three A and three B subunits, two G subunits, plus C, D, E, F, and H subunits (contains ATP catalytic site)
  • V0 domain (transmembrane): Five subunits (a, c, c’, c”, and d)

Additional isoforms of many subunits are encoded by multiple genes or alternatively spliced variants.
The encoded ATP6V0A1 protein is associated with clathrin-coated vesicles.
