Target Information

KIR2DL5A and KIR2DL5B are Killer cell immunoglobulin-like receptors (KIRs), transmembrane glycoproteins expressed by natural killer (NK) cells and subsets of T cells. The KIR genes are polymorphic and highly homologous, located on chromosome 19q13.4 within the 1 Mb leukocyte receptor complex (LRC).

KIR proteins are classified by:

  • The number of extracellular immunoglobulin domains (2D or 3D)
  • The length of the cytoplasmic domain (long [L] or short [S])

Proteins with long cytoplasmic domains contain immune tyrosine-based inhibitory motifs (ITIM) and mediate inhibitory signals upon ligand binding. Those with short domains lack ITIM motifs and instead associate with TYRO protein tyrosine kinase binding protein to transduce activating signals.

KIR proteins play a crucial role in regulating immune responses. Functionally, KIR2DL5A and KIR2DL5B act as receptors on NK cells for HLA-C alleles, preventing cell lysis by inhibiting NK cell activity.
