Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on many stem cell populations. It serves as a stem cell marker, though its expression on human hematopoietic stem cells is reversible. CD34 may function as a surface receptor involved in receptor-mediated endocytosis and regulation of adhesion, differentiation, and proliferation of hematopoietic stem cells and progenitors.
CD34 likely represents a specific developmental stage of hematopoietic cells with distinct adhesion properties. It may act as an adhesion molecule mediating stem cell attachment to the bone marrow extracellular matrix or stromal cells, and as a scaffold for lineage-specific glycans that enable binding to lectins on stromal or marrow components. Additionally, CD34 may present carbohydrate ligands to selectins. The intracellular domain can be phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
