Target Information

STK4 (MST1) is a serine/threonine kinase involved in apoptosis and chromatin condensation. It is a stress-activated, pro-apoptotic kinase that, upon caspase cleavage, enters the nucleus to induce chromatin condensation and DNA fragmentation.
STK4 is a key component of the Hippo signaling pathway, crucial for organ size control and tumor suppression by limiting proliferation and promoting apoptosis.
In this pathway, STK3/MST2 and STK4/MST1, with SAV1, activate LATS1/2 and MOB1, which phosphorylate and inactivate YAP1 and WWTR1/TAZ. Phosphorylation of YAP1 prevents its nuclear translocation, regulating genes related to proliferation, cell death, and migration.
STK3/MST2 and STK4/MST1 are essential for repressing hepatocyte proliferation, preventing activation of adult liver stem cells, and inhibiting tumor formation.
