Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
The His tag is commonly expressed at either the N- or C-terminal regions of recombinant proteins to allow isolation or purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging enables detection of recombinant proteins for which no specific antibody exists by fusing a known epitope to the protein of interest.
His-tagged proteins can be purified using metal chelate chromatography due to the strong and selective affinity between histidine residues and metal ions such as Ni²⁺.
His tag-specific antibodies are used for detection or co-immunoprecipitation of tagged proteins.
