The Oncomine Tumor Mutation Load Assay is a targeted next-generation sequencing (NGS) assay designed to assess tumor mutation load and mutation signatures through a simple, efficient workflow. It enables library construction from formalin-fixed paraffin-embedded (FFPE) tumor samples without requiring a matched normal sample, using only 10 ng of DNA per primer pool. This assay supports cancer immunotherapy research by facilitating the identification of samples most likely to respond to treatment.

The Chef-Ready Library Preparation version includes panel primers and reagents for automated library construction and normalization for up to 32 samples using the Ion Chef System.

From Sample to Answer

The assay comes with optimized informatics and visualization tools for a complete sample-to-answer workflow.

• Run metrics can be monitored with the Torrent Suite plugin.
• The Ion Reporter workflow applies custom variant calling and filtering algorithms to estimate somatic variants accurately.

A detailed report provides:

• Normalized mutation load (mutations/MB)
• Mutation signatures of somatic variants
• Percentage of mutations consistent with UV damage, tobacco smoke damage, deamination, and specific substitutions
