Product Specific Information

The OKT8 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer. CD8a is found on thymocytes, T cells, and NK cells, binding MHC class I and associating with kinase p56lck to regulate T-cell development and activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells (5 µL/test, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails or master mixes.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD8 is a type I transmembrane glycoprotein functioning as a co-receptor for MHC class I molecules. It is expressed on T cells, NK cells, monocytes, and dendritic cells. CD8α binds MHC-I, enabling signal transduction and CTL activation. Activated CTLs play essential roles in pathogen and tumor cell clearance. Differentiation of CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
