Target Information
Neurogenic locus notch homolog protein 2 (NOTCH2) is a highly conserved protein in the Notch signaling pathway. It is a type I transmembrane receptor with an extracellular domain containing multiple epidermal growth factor-like (EGF) repeats and an intracellular domain with multiple distinct domains. NOTCH2 interacts with membrane-bound ligands such as Jagged1, Jagged2, and Delta1 to regulate cell-fate determination.
Defects in NOTCH2 are associated with Alagille syndrome type 2 (ALGS2), an autosomal dominant multisystem disorder characterized by hepatic bile duct paucity, cholestasis, and various cardiac, skeletal, and ophthalmologic abnormalities. Additional features may include characteristic facial morphology and occasional renal or vascular involvement.
