Product Specific Information
The W6/32 monoclonal antibody reacts with the human major histocompatibility complex (MHC) class I, HLA-A, B, and C. These antigens, associated with beta 2-microglobulin, are expressed on all human nucleated cells and play a central role in cell-mediated immune response and tumor surveillance. The W6/32 clone recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity is observed in baboon, rhesus, and cynomolgus monkey.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells at 5 µL (1 µg) per test.
A test corresponds to staining a cell sample in a final volume of 100 µL (typically 10⁵–10⁸ cells/test).
Excitation: 488 nm; Emission: 520 nm; Laser: Blue.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
HLA-class I major histocompatibility (MHC) antigens are intrinsic membrane glycoproteins expressed on nucleated cells and noncovalently associated with beta2-microglobulin. They present foreign determinants for recognition by cytotoxic T cells, playing key roles in antiviral and antitumor defense. Human HLA-class I antigens include HLA-A, HLA-B, and HLA-C molecules.
HLA and MHC antibodies are essential in Immunopeptidomics, aiding identification and characterization of neoantigens via high-performance liquid chromatography coupled to tandem mass spectrometry.
