Target Information
BPTF (bromodomain and PHD domain transcription factor) is the largest subunit of the ATP-dependent chromatin-remodelling complex, NURF (nucleosome remodelling factor). NURF catalyzes ATP-dependent nucleosome sliding and facilitates transcription. BPTF recognizes histone H3 tails that are tri-methylated at K4, marking transcriptional start sites of most active genes. It also binds to H3 di-methylated at K4.
BPTF plays a key role in early mouse embryo development, possibly through regulation of the Smad transcription factor pathway. It is expressed at low levels in adult brain and spinal cord but at higher levels in neurodegenerative diseases. Found in amyloid-containing plaques of Alzheimer’s disease brains, and abundantly expressed in fetal brain. Present throughout gray and white matter of the developing spinal cord at 18–22 gestational weeks. Tissue specificity: ubiquitously expressed with highest levels in testis; also present in kidney, liver, and brain, with highest levels in motor cortex.
