Target Information
FOXO1 (FKHR, ForKHead Box O1)는 전사인자(forkhead homeotic gene family)의 하위 패밀리에 속하는 단백질로, 핵 수용체 활성의 공동활성자(coactivator) 또는 공동억제자(corepressor)로 작용할 수 있습니다.
FOXO1과 PAX3의 결합은 alveolar rhabdomyosarcoma와 관련이 있으며, FOXO1은 자가포식(autophagy), 근육 성장 및 분화 조절, 항암 활성과 관련된 중요한 인자로 보고됩니다.
또한, FOXO1은 인슐린 신호 전달의 주요 표적 단백질로, 산화 스트레스에 대한 대사 항상성 조절에 관여합니다.
FOXO1은 인슐린 반응 요소(IRE, 5'-TT[G/A]TTTTG-3') 및 Daf-16 결합 요소(DBE, 5'-TT[G/A]TTTAC-3')에 결합하며, 산화환원 균형, 조골세포 수 조절, 골량 조절에 중요한 역할을 합니다.
간세포에서는 PPARGC1A와 함께 작용하여 IGFBP1, G6PC, PCK1 등의 유전자 발현을 촉진함으로써 포도당신생합성을 유도합니다.
또한 FOXO1은 CDK1, PKB/AKT1, SKT4/MST1 등의 신호 하류에서 세포 사멸 조절자로 작용합니다.
