Target Information
Sin1 (MAPKAP1) was initially identified as the human homolog of S. pombe SIN1. It interacts with MEKK2, a component of the MAPK signaling network, and is thought to prevent MEKK2 activation and dimerization by forming a complex with inactive MEKK2. This interaction blocks activation of JNK1/2, ERK1/2, p38, and ERK5 MAPKs.
MAPKAP1 also plays a role in TOR signaling, regulating cell growth and proliferation in response to nutrients and growth factors. It maintains the TOR/rictor complex, enabling specific phosphorylation of Akt, linking growth factor–PI3K signaling with nutrient-regulated TOR signaling. Multiple alternatively spliced isoforms of MAPKAP1 have been identified.
