BRAF는 세포막에서 핵으로의 유사분열 신호 전달에 관여하며, 인간 암에서 체세포 점 돌연변이에 의해 활성화됩니다. 또한 해마 뉴런의 시냅스 후 반응에 관여하는 것으로 알려져 있습니다. 이 단백질은 MAP kinase/ERKs 신호 경로를 조절하여 세포 분열, 분화 및 분비에 영향을 미칩니다.
B-Raf의 돌연변이는 비호지킨 림프종, 대장암, 악성 흑색종, 갑상선암, 비소세포폐암 및 폐선암 등 다양한 암과 관련이 있습니다.
또한 X 염색체에 위치한 B-Raf의 유사유전자(pseudogene)가 확인되었습니다.
