Description:
The WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein and a member of the Siglec (sialic acid-binding Ig superfamily lectin) family. CD33 is expressed in the hematopoietic compartment, including monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Applications Reported:
This antibody is validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Target Information

CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit phosphatases SHP-1 and SHP-2, regulating signal transduction and receptor removal.
CD33 is expressed on myelomonocytic lineage cells, binds sialic acid residues on glycoproteins, and serves as a therapeutic target for acute myeloid leukemia. It is also found on granulocyte and macrophage precursors in bone marrow and peripheral monocytes, making it a useful marker for distinguishing myelogenous leukemia cells from other lineages.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
