TPMT (Thiopurine S-methyltransferase) catalyzes the S-methylation of thiopurine drugs such as 6-mercaptopurine.
Defects in TPMT cause TPMT deficiency, affecting metabolism of thiopurine drugs used as immunosuppressants or cytotoxic agents.
Patients with reduced TPMT activity risk toxicity under standard dosing due to genetic variation at the TPMT locus.
