Target Information

Inositol 1,4,5-trisphosphate (IP3) acts as a second messenger for various growth factors, hormones, and neurotransmitters. Upon binding to IP3R, IP3 triggers the release of intracellular calcium to the cytosol. Functional IP3R is a homo- or heterotetramer of approximately 240 kDa glycoprotein subunits. IP3R is structurally and functionally related to the ryanodine receptor, another intracellular calcium-release channel. Both receptors can associate with the immunophilin protein FKBP12.

Phosphorylation of IP3R by PKC enhances IP3-mediated calcium release, while dephosphorylation by calcineurin reduces it, leading to calcium oscillations. Mammalian IP3R subunits are encoded by three distinct genes with differential expression:

  • IP3R-I: heart, liver, kidney, ovary, cerebellar Purkinje neurons
  • IP3R-II: predominantly in brain
  • IP3R-III: pancreatic islets, kidney, gastrointestinal tract
