Product Specific Information

The V9 monoclonal antibody recognizes human Vimentin, a 57 kDa protein functioning as a structural component of intermediate filaments. Vimentin is expressed in mesenchymal-derived cells and specific populations such as radial glia, immature glial cells, and pancreatic precursor cells. It serves as a marker of cardiac differentiation and is gradually replaced by neurofilaments in neural cells.

This antibody also recognizes canine, rat, and chicken vimentin but does not recognize mouse vimentin.

Applications Reported: Immunocytochemistry and immunohistochemical staining of frozen (IHC-F) and formalin-fixed paraffin-embedded tissue sections (IHC-P).

Applications Tested: Tested on fixed and permeabilized C6 cells at ≤5 µg/mL. Optimal performance should be determined by titration.

Filter Recommendation: For eFluor™ 570 conjugates, use filters capturing the 570 nm emission wavelength (e.g., Excitation 545/25, 565LP, Emission 605/70). Alexa Fluor™ 555 or TRITC filters are acceptable.

Target Information

Vimentin is a developmentally regulated intermediate filament protein (IFP) found in mesenchymal cells. It is involved in intracellular protein transport between the nucleus and plasma membrane. Expressed with desmin during early development, vimentin is later replaced by tissue-specific IFPs. It contributes to steroid synthesis regulation by acting as a storage network for cholesterol-containing lipid droplets. Phosphorylation by protein kinase leads to filament breakdown and cytoskeletal changes that facilitate cholesterol transport to mitochondria, enhancing steroid synthesis.
