Target Information
Members of the TNF superfamily regulate immune responses and induce apoptosis. Two novel members, BAFF (also known as BLyS, TALL-1, THANK, zTNF4) and APRIL (TALL-2), have been identified.
BAFF acts as a B cell activator, promoting B cell proliferation and immunoglobulin secretion, while APRIL regulates both immunological and non-immunological cell growth.
Three receptors—BCMA, TACI, and BAFF-R—bind BAFF and APRIL. TACI, like BCMA, interacts with both ligands and plays a key role in regulating humoral immune responses, activating NF-κB and JNK pathways, and contributing to B cell–associated autoimmune diseases.
