This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (Fc gamma RIII).
CD16 exists as two isoforms, FcγRIIIA and FcγRIIIB. FcγRIIIA is expressed on monocytes, macrophages, and NK cells, while FcγRIIIB is found on neutrophils. Expression patterns are similar in non-human primates, with some species-specific variations.

Binding of IgG induces signal transduction, leading to ADCC, phagocytosis, cytokine release, and antigen presentation.
3G8 recognizes the same epitope as CB16 but differs from B73.1.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for flow cytometry on human peripheral blood cells.
Use 5 µL (0.2 µg)/test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
Add 5 µL per sample to antibody cocktails before combining with cells.

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures designed to minimize spillover and spread.

Target Information

CD16 encodes the Fcγ receptor that binds the Fc portion of IgG, mediating immune complex clearance, ADCC, and viral infection enhancement.
The FCGR3A gene encodes the membrane-bound form on NK cells, while FCGR3B encodes the PI-linked form on neutrophils.
Mutations are associated with Immunodeficiency 20, Herpes Zoster, and autoimmune conditions such as systemic lupus erythematosus.
