Target Information
Sclerostin은 주로 골세포(osteocytes)에서 분비되는 당단백질로, Cerberus/DAN 계열의 BMP 길항제에 속하며 Wnt 신호전달 경로의 내인성 조절자로 작용합니다. 이 단백질은 골 항상성의 억제적 조절자 역할을 하며, 뼈, 골수, 연골, 신장, 간 등에서도 발현됩니다. 또한 비대성 연골세포 및 cementocyte에서도 생성될 수 있습니다.
Sclerostin은 DKK-1 및 DKK-4와 유사하게 Wnt/β-catenin 신호 경로에서 LRP5 및 LRP6과 결합하여 억제 복합체를 형성함으로써 Wnt 신호를 억제합니다. 또한 LRP4의 세포외 도메인과 직접 상호작용하여 Wnt 신호 억제를 매개하며, RANKL 발현을 증가시켜 파골세포 활성화를 촉진할 수 있습니다.
Sclerostin의 이러한 조절 기능은 sclerosteosis 및 van Buchem disease(VBD)와 같은 희귀 유전성 골 이형성 질환에서 그 중요성이 강조되며, 이들 질환은 sclerostin 결핍으로 인한 과도한 골 형성과 과광화로 특징지어집니다.
