Target Information

ApoE belongs to a group of proteins that bind reversibly with lipoprotein and play an important role in lipid metabolism. These proteins facilitate lipid solubilization, maintain lipoprotein structural integrity, serve as ligands for lipoprotein receptors, and regulate enzymes involved in lipid metabolism.
ApoE is produced mainly in the liver and brain and is a constituent of all plasma lipoproteins. Its interaction with ApoE receptors enables uptake of chylomicron remnants by liver cells, essential for normal lipid metabolism. ApoE also binds with the LDL receptor (apo B/E).
Defects in ApoE cause hyperlipoproteinemia type III. ApoE exists in three major isoforms—E2, E3, and E4—differing by a single amino acid substitution. Compared with E3 and E4, E2 has the lowest receptor binding affinity. E2 allele carriers show lower total cholesterol, LDL cholesterol, and non-HDL cholesterol, with increased ApoE levels.
