Target Information
CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa depending on phosphorylation state).
It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells.
CD20 regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
It associates with lipid rafts in a dynamic manner depending on extracellular triggering and BCR aggregation.
After receptor ligation, CD20 and BCR colocalize and then dissociate before BCR endocytosis, while CD20 remains on the cell surface.
CD20 is a key therapeutic target for B-cell depletion in malignancies, as it is highly expressed and not internalized or shed after antibody treatment.
Diseases associated with CD20 dysfunction include MS4A1-related common variable immune deficiency.
