Target Information

Mitochondrial transcription factor A (TFAM) is a crucial nuclear-encoded protein transported to mitochondria, where it maintains and regulates mitochondrial DNA (mtDNA) replication and transcription. TFAM interacts with LC3 via its LC3 interacting region (LIR) motif, playing an essential role in autophagy and inflammation control by eliminating leaked mtDNA. TFAM levels influence mitochondrial function; high expression can repress mtDNA transcription in skeletal muscle, while balanced TFAM-to-mtDNA ratios preserve oxidative phosphorylation capacity in cardiac tissue. Dysregulation of TFAM is associated with neurodegeneration and diseases related to mtDNA stress.
