Target Information

SIGIRR (single Ig IL-1R-related molecule) is a novel member of the interleukin 1 receptor (IL-1R) superfamily. Although it shares conserved motifs with the IL-1R and Toll superfamily, it is structurally and functionally distinct. SIGIRR has a single Ig domain in its extracellular region, unlike the three Ig folds in IL-1R family members. Its long cytoplasmic domain resembles that of Drosophila Toll, but its peptide sequence is more closely related to IL-1RI. A chimera with the SIGIRR intracellular domain fused to the IL-1R extracellular domain did not activate NF-κB, unlike other IL-1R-related fusion proteins. Phylogenetic data suggest that the plasma membrane protein SIGIRR is a functional relative of MyD88 in mammals.
