Target Information

Phosphofructokinase (PFK) is a major regulatory enzyme involved in sugar metabolism and energy production in all cells. In humans, three structural loci control PFK subunits: M (muscle), L (liver), and P (platelet). These subunits form various tetramers, with muscle and liver PFKs being homotetramers (M₄ and L₄). Red blood cells express all five isozymes (M₄, M₃L, M₂L₂, ML₃, L₄). PFK is allosterically activated by ADP, AMP, or fructose bisphosphate and inhibited by ATP or citrate. It catalyzes the key regulatory step in glycolysis. PFK deficiency can cause episodic illness with dark-colored urine following strenuous exercise, and elevated PFK levels are observed in individuals with Down syndrome.
