Specificity Comments:
Recognizes a 130–140 kDa protein identified as CD22 (also known as BL-CAM). Expression is restricted to normal and neoplastic B cells, absent in other hematopoietic cell types.
CD22 appears in the cytoplasm of pro-B and pre-B cells and on the surface of mature IgD+ B cells. Not expressed by plasma cells. Highly expressed in follicular mantle and marginal zone B cells; weak in germinal center B cells.
CD22 belongs to the immunoglobulin superfamily, acts as an adhesion receptor for sialic acid-bearing ligands on erythrocytes and leukocytes, and associates with tyrosine kinases involved in B-cell activation and signal transduction.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and membrane of B-lymphocytes.
It appears early in B-cell differentiation, similar to CD19, and is useful for phenotyping mature leukemias.
Strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma; negative in classical Hodgkin’s lymphoma.
CD22 binds preferentially to α2,6-linked sialic acid, with recognition modulated by cis interactions.
Upon ligand-induced tyrosine phosphorylation, CD22 regulates B-cell receptor signaling through interactions with Src family kinases and phosphatases.
