Target Information
The human UNG gene encodes both nuclear (UNG2) and mitochondrial (UNG1) forms of uracil-DNA glycosylase. These isoforms are generated by alternative splicing and the use of two differentially regulated promoters, PA and PB.
UNG1 and UNG2 cDNAs are similar in size (2061 and 2058 bp, respectively), making them indistinguishable in gel electrophoresis.
Nuclear UNG2 differs from mitochondrial UNG1 by 44 amino acids at the N-terminal region, which is not required for catalytic activity.
A major role of UNG gene products is to repair mutagenic U:G mispairs caused by cytosine deamination. For example, UNG2 removes misincorporated dUMP residues. Expression levels of UNG1 and UNG2 vary by cell and tissue type and are regulated by the cell cycle.
Expression and activity of UNG are generally higher in proliferating compared to non-proliferating cells.
UNG1 is a 304 amino acid protein, while UNG2 consists of 313 amino acids.
