Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor (~90 kDa) has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain.
AR is a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.
The AR signaling pathway is critical for the development and function of male reproductive organs (e.g., prostate, epididymis) and also plays roles in nonreproductive tissues such as muscle, hair follicles, and brain.
Androgen receptor is a phosphoprotein that regulates MAP kinase signaling; inhibition of the MEK1/2 pathway correlates with changes in AR phosphorylation state.
Abnormalities in AR signaling are associated with diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in this gene can result in complete androgen insensitivity (CAIS).
