Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either the N- or C-terminal regions of recombinant proteins to enable purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging allows detection of recombinant proteins lacking specific antibodies by fusing a known epitope to the target protein. His-tagged proteins can be purified using Ni²⁺-NTA adsorbents due to the strong and selective chelation between histidine clusters and nickel ions.
His tag-specific antibodies facilitate detection and co-immunoprecipitation of tagged proteins.
