Target Information
This gene is one of seven beta-1,4-galactosyltransferase (beta4GalT) genes. They encode type II membrane-bound glycoproteins with specificity for UDP-galactose, transferring galactose in a beta1,4 linkage to acceptor sugars such as GlcNAc, Glc, and Xyl. Each beta4GalT has distinct roles in glycoconjugate and saccharide biosynthesis.
As type II membrane proteins, they possess an N-terminal hydrophobic signal sequence that directs localization to the Golgi apparatus and serves as a transmembrane anchor. The beta4GalTs form four groups based on sequence similarity:
- beta4GalT1 & beta4GalT2
- beta4GalT3 & beta4GalT4
- beta4GalT5 & beta4GalT6
- beta4GalT7
B4GALT1 is unique among these genes as it participates in both glycoconjugate and lactose biosynthesis. It adds galactose to N-acetylglucosamine residues on glycoprotein carbohydrate chains and, in lactating mammary tissues, forms a heterodimer with alpha-lactalbumin to catalyze UDP-galactose + D-glucose ⇔ UDP + lactose. The two enzymatic forms arise from alternative transcription initiation and post-translational processing.
