SIRT5는 Sirtuins(Sir2-like proteins) 계열에 속하는 인간 단백질로, 원핵 및 진핵생물에 존재합니다. 모든 Sir2-like 단백질은 박테리아부터 인간까지 보존된 시퀀스 모티프를 포함하는 sirtuin core domain을 가지고 있습니다. 박테리아, 효모, 포유류의 sirtuins은 NAD를 대사하고 mono-ADP-ribosyltransferase로 작용할 수 있습니다. 효소 기능은 완전히 규명되지 않았지만, 최근 연구에서 Sir2가 NAD 대사 및 histone 탈아세틸화에 관여함을 보여주며, NAD와 acetylated histone 사이의 상호작용을 통한 활성화 메커니즘 가능성을 시사합니다.
