Target Information
The human AF9 gene is one of the most common fusion partner genes with the ALL1 (MLL) gene at 11q23, resulting in t(9;11)(p22;q23) translocation. The AF9 gene spans over 100 kb and includes two patient breakpoint cluster regions (BCRs):
- BCR1 within intron 4 (site A)
- BCR2 spanning introns 7 and 8 (site B)
AF9 contains structural elements such as:
- In vivo DNA topo II cleavage site
- In vitro DNase I hypersensitive site in intron 7 (BCR2)
- Two scaffold associated regions (SARs): SAR1 (intron 4) and SAR2 (exons 5–7)
These regions share structural similarities with the MLL breakpoint cluster region (BCR). A DNA breakage and repair model for nonhomologous recombination between MLL and AF9 has been proposed.
