The Pierce Bradford Plus Protein Assay is a ready-to-use, reducing agent-compatible, improved Bradford assay reagent for quick measurement of total protein concentration compared to a protein standard. This reagent provides increased linearity and half the protein-to-protein variability of other commercial Bradford assay formulations.

사용 방법

단일 ready-to-use 용액으로 시료와 표준 단백질에 동일한 부피로 첨가 후 혼합하여 595 nm에서 흡광도를 측정합니다. 시험관 또는 마이크로플레이트 형식으로 수행 가능하며, 대부분의 염, 용매, 버퍼, 티올, 환원제 및 금속 킬레이트제와 호환됩니다.

원리 (How the Bradford Plus Assay Detects Protein)

Coomassie G-250 염료는 단백질과 결합 시 스펙트럼이 465 nm(적갈색)에서 610 nm(청색)으로 이동합니다. 595 nm에서 두 형태 간의 차이가 가장 커 측정에 최적입니다. 575–615 nm 범위에서도 측정 가능하나, 양 극단에서는 약 10% 감쇄가 발생합니다.
색상 발현은 주로 아르기닌, 라이신, 히스티딘 등 염기성 아미노산과의 결합에 의해 발생하며, 반데르발스 및 소수성 상호작용도 관여합니다. 분자량 3,000달톤 이하의 펩타이드나 단백질은 염료와 결합하지 않아 측정되지 않습니다.
