Target Information
In mammals, several isoforms of p21-Activated Protein Kinases (PAKs) have been identified:
- α-PAK (PAK-1) and β-PAK (PAK-3) are primarily brain-specific.
- γ-PAK (PAK-2) is expressed ubiquitously.
Mutations in the PAK-3 gene are linked to X-linked mental retardation, and PAK-3 is a key regulator of synapse formation and hippocampal plasticity. PAK-3 also plays a crucial role in regulating cell shape, motility, and apoptosis. Autophosphorylation of Thr402 is essential for PAK activation.
