Target Information
Na⁺/H⁺ exchangers (NHE) are plasma membrane intrinsic proteins that mediate the exchange of Na⁺ and H⁺ ions in various tissues. They play a critical role in:
- Regulation of intracellular pH (pHi)
- Trans-cellular absorption of Na⁺
- Cell volume regulation
- Cell proliferation
These functions are associated with several pathophysiological states such as hypertension, organ growth, hypertrophy, cancer regression, and renal or intestinal disorders.
At least seven NHE isoforms (NHE1–7) have been identified, sharing similar primary structures with 10–12 predicted transmembrane domains. The C-terminal domain is predicted to be intracellular.
NHE7 (human, 725 aa, chromosome Xp11.4) is ubiquitously expressed and predominantly localized to the trans-Golgi network. It mediates the influx of Na⁺ or K⁺ in exchange for H⁺ and shows approximately 70% similarity to NHE6 but only about 25% homology with other NHE isoforms.
