Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L, also known as lymphocyte function-associated antigen-1 (LFA-1) alpha chain. LFA-1, formed by non-covalent association of CD11a with CD18 (integrin beta 2), serves as an important adhesion molecule involved in lymphocyte and granulocyte function. CD11a is expressed by all leukocytes.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse splenocytes at ≤0.4 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
  Optimize antibody titration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails prior to mixing with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for reduced spillover and spread.

Note:
Internal testing shows non-specific B-cell staining in SJL mice. Not observed in BALB/c or C57BL/6 strains.

Target Information

CD11a (LFA-1 alpha, ITGAL) forms part of the leukocyte function-associated antigen 1 (LFA-1, αLβ2 integrin) with CD18. It mediates leukocyte adhesion and costimulatory signaling via ICAM-1, -2, and -3. CD11a expression is ubiquitous on leukocytes and transitions from low- to high-affinity states upon stimulation. It plays key roles in immune cell interactions, migration, differentiation, and T-cell effector function.

Clinical Relevance:

• Target in inflammatory disease therapies (e.g., Efalizumab/Raptiva).
• Simvastatin binding to CD11a modulates immune activity.
• Associated diseases: benign cephalic histiocytosis, leukocyte adhesion deficiency.
