Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor functions as a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA.
ER can regulate gene transcription positively or negatively through recruitment of coactivator or corepressor proteins.
Two forms of estrogen receptor exist — alpha (ESR1) and beta (ESR2) — encoded by separate genes. Alternative RNA splicing produces at least four known ERα isoforms (66, 53, 47, and 35 kDa).
Estrogen receptors are widely expressed in various tissues and are essential for sexual development, reproductive function, and bone physiology. They are also implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
