Target Information
BRCC36 forms a holoenzyme complex with BRCA1, BRCA2, RAD51, and BRCC45, possessing ubiquitin E3 ligase activity. Aberrant BRCC36 levels are found in sporadic breast tumors. Depletion of BRCC36 or BRCC45 by siRNA increases sensitivity to ionizing radiation and causes G2/M checkpoint defects, suggesting BRCC36 enhances cellular survival following DNA damage. BRCC36 is essential for radiation-induced BRCA1 phosphorylation and nuclear foci formation, indicating potential as a therapeutic target for radiation-resistant breast tumors. At least two isoforms of BRCC36 are known.
