Target Information
Specialized cells at the midline, which separates the left and right halves of the CNS, play key roles in directing growth cone behavior. In vertebrates, insects, and C. elegans, midline cells produce guidance cues such as nectins and slit, acting as attractants and repellents, respectively. Robo proteins serve as axon guidance receptors within the immunoglobulin superfamily, conserved from fruit flies to mammals. Robo1 acts as a receptor for the repellent Slit and functions in a cell-autonomous manner. Non-crossing axons express high levels of Robo, while crossing axons express low levels before reaching the midline and high levels after crossing. Robo1 and Robo2 are human homologs of the Drosophila protein Roundabout.
