PDZ-GEF1 (RAPGEF2)는 cAMP 의존적 방식으로 Rap 및 Ras 계열의 작은 GTPase를 활성화시키는 guanine nucleotide exchange factor(GEF)로 작용합니다. 세포 표면 수용체와 Rap/Ras GTPase 간의 신호 전달을 연결하며, Rap1-GTP와 직접 결합하여 Rap1 매개 신호를 증폭시킵니다. HRAS에 대해서는 약한 활성을 보입니다.
RAPGEF2가 cAMP 및 cGMP에 결합하는지 여부는 논쟁 중이며, β1-아드레날린 수용체(ADRB1)와의 결합을 통해 G(s)-alpha 신호 경로를 통한 Ras 활성화를 유도합니다. 또한 cAMP 유도 Ras 및 Erk1/2 신호 경로에 관여하며, 이는 멜라닌 생성의 장기 억제와 관련됩니다. 신경세포의 신장 및 뇌 발달 과정에서 축삭 연결 형성에도 관여하며, 혈관 형성과 내피세포 간 접합의 안정성 조절에도 역할을 합니다.
