Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a transcription factor essential for thymic development and Th2 differentiation. Gata-3 expression is observed in the CNS, skin, mammary glands, and kidney during embryonic development, and is critical for T-cell commitment and survival. In the thymus, expression occurs mainly in CD4 single-positive cells.

Alternative splice variants (50 and 45 kDa) are recognized by this antibody.
Staining requires the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Cross-reactivity in rhesus monkeys has been reported.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and tested for intracellular staining of mouse thymocytes using Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended usage: 5 µL (0.5 µg)/test.

Brilliant Violet™ 786 (BV786) emits at 786 nm and is compatible with cytometers equipped with a 405 nm laser. Use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) when combining multiple polymer dye-conjugated antibodies to minimize non-specific interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
Note: BV786 is not compatible with methanol-based fixation.

Target Information

GATA3 is a transcription factor that binds to enhancer elements of T-cell receptor (TCR) genes (alpha, beta, gamma, delta) and regulates their expression. It contains a zinc finger domain and is abundantly expressed in T-lymphocytes, playing a crucial role in TCR gene activation. The human gene spans 17 kb with six exons, encoding two zinc fingers highly conserved with GATA1.
