Antibody detects endogenous levels of total NLK.
Target Information
NLK is a serine/threonine kinase involved in signal transduction and cellular communication. It acts as a positive effector of the non-canonical Wnt signaling pathway, functioning downstream of WNT5A, MAP3K7/TAK1, and HIPK2. Activation leads to phosphorylation of SETDB1, interaction with PPARG, and transcriptional silencing of PPARG target genes, thereby inhibiting adipogenesis and promoting osteoblastogenesis in mesenchymal stem cells. NLK negatively regulates canonical Wnt/beta-catenin signaling by phosphorylating TCF7L2/TCF4 and LEF1, leading to complex dissociation and proteolysis. It also negatively regulates Notch and MYB signaling pathways by phosphorylating NOTCH1 and MYB family transcription factors.
