Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except mature erythrocytes and platelets. Its cytoplasmic domain has tyrosine phosphatase activity and plays a key role in lymphocyte activation.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested: Flow cytometry of mouse bone marrow cells (≤0.4 µg/test)

Usage Recommendations:

• Optimize antibody titration for best performance.
• Avoid DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It is absent in non-hematopoietic tissues and cell lines. CD45 regulates Src-family kinases, lymphocyte development, and antigen signaling, and influences integrin-mediated signaling and apoptosis. Multiple isoforms arise from alternative splicing, with expression varying by cell type and physiological state.
