This product is diluted and ready-to-use.
Recommended positive control tissues include epithelial cells (tonsil, prostate, pancreas, colon, thyroid, etc.), though other tissues may also be suitable.

The antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue for manual qualitative IHC testing. It should be used after tumor diagnosis by conventional histopathology.

Cytokeratin 8 (CK8) belongs to the type II (basic) subfamily of high molecular weight cytokeratins and pairs with cytokeratin 18 (CK18). This monoclonal antibody recognizes all simple epithelia, including glandular epithelium (thyroid, breast, GI tract, respiratory and urogenital tracts). All adenocarcinomas and most squamous carcinomas are positive, while keratinizing squamous carcinomas are generally negative. It is useful for detecting Paget cells, which stain due to the presence of keratin 18.

Antibody is used on formalin-fixed, paraffin-embedded sections. Pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens through sequential application of a primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, followed by washing steps. The resulting chromogen reaction is observed under a light microscope to aid in differential diagnosis.

A positive tissue control must accompany every staining procedure. This control should contain both positive and negative components. External positive control tissues should be freshly prepared autopsy, biopsy, or surgical specimens fixed and processed similarly to patient samples. Weakly positive tissues are preferred for quality control to detect minor reagent degradation or sensitivity changes.

Internal or external negative controls should be used according to institutional guidelines. Negative control sites should show absence of specific staining and minimal background. If specific staining appears in negative controls, the test results are invalid.

Target Information

Keratin 8 belongs to the type B (basic) subfamily of high molecular weight keratins and pairs with cytokeratin 18 (type A; acidic subfamily of low molecular weight keratins).
