Recommended positive control tissue: ZMTB-1, though other tissues may also be used.
Intended for professional laboratory use in detecting Cyclin E protein in formalin-fixed, paraffin-embedded tissue by manual qualitative IHC testing.
Cyclin E is a member of the conserved cyclin family, regulating CDK kinases and controlling the G1/S transition of the cell cycle. Overexpression of Cyclin E has been observed in various tumors, contributing to chromosomal instability and tumorigenesis.
Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended. Immunohistochemical staining visualizes antigens via sequential antibody and enzyme reactions, producing a visible chromogenic product for microscopic interpretation.
A positive tissue control must be included in every staining procedure. Weakly positive tissues are ideal to monitor antibody sensitivity and staining consistency.
Negative control tissues should confirm absence of specific staining and help detect non-specific background signals.
Target Information
Cyclin E regulates CDK2 activity and is essential for cell cycle G1/S transition. It accumulates at the G1-S boundary and degrades during S phase. Overexpression is linked to tumorigenesis due to chromosomal instability.
Cyclin E associates with NPAT protein, influencing histone gene expression and promoting cell-cycle progression in absence of pRB.
Two alternatively spliced transcript variants encode distinct isoforms; additional splice variants have been reported but not fully characterized.
