Target Information

Protein Z (PROZ) is a vitamin K–dependent plasma glycoprotein (~62 kDa, 396 amino acids) involved in the coagulation cascade. It contains four domains: a Gla-rich region, two EGF-like domains, and a trypsin-like domain. Although structurally related to serine proteases (factors VII, IX, X, and protein C), it lacks catalytic activity. Protein Z enhances the degradation of factor Xa by protein Z–related protease inhibitor (ZPI), accelerating the reaction up to 1000-fold. Deficiency has been variably associated with thrombosis or bleeding tendencies.
