Target Information
BACE1 (beta-secretase)는 Alzheimer’s disease(AD)에서 발견되는 amyloid beta-peptide(Abeta) 생성에 관여하는 핵심 효소입니다. 일부 조기 발병형 가족성 AD는 APP의 ‘Swedish’ 돌연변이로 인해 BACE1에 의해 단백질 절단이 증가함으로써 발생합니다. 이러한 유전적 및 병리학적 근거를 바탕으로 BACE1 및 gamma-secretase 억제를 통한 치료 접근이 연구되고 있습니다. APP는 먼저 b-secretase에 의해 절단되어 용해성 단백질과 막 결합형 C99 단편을 생성하며, C99는 g-secretase의 기질로 작용해 4 kDa amyloid-beta 펩타이드를 생성합니다. 이 펩타이드는 AD 환자의 뇌에 축적됩니다. BACE1은 beta-site APP cleaving enzyme(BACE) 및 aspartyl protease 2(Asp2)로도 알려진 새로운 막관통성 아스파르트산 프로테아제입니다.
