Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either the N- or C-terminal regions of recombinant proteins to enable isolation or purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging involves fusing a known epitope to a recombinant protein using genetic engineering, allowing detection of proteins for which no specific antibody exists. His-tagged proteins are purified via metal chelate chromatography due to the strong and selective affinity of histidine clusters for metal ions such as Ni²⁺. This enables purification to homogeneity using Ni²⁺-NTA adsorbents.
His tag-specific antibodies facilitate detection and co-immunoprecipitation of tagged proteins.
